Braga, Karina de Queiroz; Coimbra, Mairon César; Castro, Ana Hortência Fonsêca.
A protocol for the in vitro germination and callus induction in Pyrostegia venusta, a medicinal plant species from the Brazilian savannah (Cerrado) is provided. The establishment of cultures of P. venusta was originally obtained from seeds germinated in vitro and induced callus directly from leaf explants of seedlings. Seeds were germinated on MS and WPM media containing 100 and 50% of salt concentration, supplemented with 30 g L-1 sucrose. Callus induction consisted of the inoculation of leaf segments on MS medium plus 2,4-D or BAP in the presence or absence of light. The germination percentage averaged 85% and the aerial parts and roots of seedlings obtained in WPM with 50 and 100% of salt concentration showed elevated contents of total phenolic...

CASTRO,ANA HORTÊNCIA FONSÊCA; YOUNG,MARIA CLÁUDIA MARX; ALVARENGA,AMAURI ALVES DE; ALVES,JOSÉ DONIZETI.
In order to evaluate the influence of photoperiod on the accumulation of allantoin in comfrey (Symphytum officinale L.), a well known medicinal plant, an experiment was conducted during July and August, 1998. Cuttings obtained from 90 day old plants were submitted to four photoperiods (8, 12, 16 and 20 hours). After 60 days, allantoin content in roots and rhizomes was evaluated. The results showed that increases in photoperiod promoted an increment in the average content of allantoin in roots (0.06%, 0.303%, 1.213% and 4.78%). On the other hand, in rhizomes, allantoin accumulation decreased (9.65%, 7.14%, 0.55%) when the photoperiod was increased from 8 to 12 and 16 hours, respectively, stabilizing on a 20-hour photoperiod (0.53%). Plants cultivated under...

Azevedo,C.R.; Maciel,F.M.; Silva,L.B.; Ferreira,A.T.S.; da Cunha,M.; Machado,O.L.T.; Fernandes,K.V.S.; Oliveira,A.E.A.; Xavier-Filho,J..
Evidence based on immunological cross-reactivity and anti-diabetic properties has suggested the presence of insulin-like peptides in plants. The objective of the present study was to investigate the presence of insulin-like proteins in the leaves of Bauhinia variegata ("pata-de-vaca", "mororó"), a plant widely utilized in popular medicine as an anti-diabetic agent. We show that an insulin-like protein was present in the leaves of this plant. A chloroplast protein with a molecular mass similar to that of bovine insulin was extracted from 2-mm thick 15% SDS-PAGE gels and fractionated with a 2 x 24 cm Sephadex G-50 column. The activity of this insulin-like protein (0.48 mg/mL) on serum glucose levels of four-week-old Swiss albino (CF1) diabetic mice was...

Shahanaz Beegum,A; Poulose Martin,Kottackal; Zhang,Chun-Lai; Nishitha,I. K.; Ligimol,; Slater,Adrian; Madhusoodanan,P. V.
Camptothecin (CPT) analogues and derivatives serve as a novel class of effective anticancer agents that exert their action against DNA topoisomerase I. This paper presents procedures for the rapid, high frequency regeneration of a camptothecin producing plant, Ophiorrhiza prostrata D. Don from leaf and internode explants via shoot organogenesis. The concentrations of plant growth regulators and explant types exhibited discrete roles in the efficacy of shoot induction. N6-benzyladenine (BA) was the most effective cytokinin for the induction of shoots. Murashige and Skoog (MS) medium with 8.87 µM BA and 2.46 µM indole-3-butyric acid (IBA) yielded the highest number of shoots from leaf and internode explants (76.0 and 90.8 shoots respectively). In the case of...
